Description88 TAg Stable Y-LIZ DNA Polymerase-β Knockout Mouse Embryonic Fibroblast Cell Line carries the lambda-LIZ transgene (Y-LIZ), a shuttle vector which enables for mutation analysis. The 92 TAg is the wild type cell line to the polymerase beta knock out cells available at abm (Cat. No. T3169, 92 TAg Stable Y-LIZ Wildtype Mouse Embryonic Fibroblast Cell Line). Both the wild type and the polymerase beta knock out cells are valuable tools to identify mutations due to the stable integration of Y-LIZ and further analysis on repair pathways.

With the absence of the polymerase-β, the cells are unable to efficiently repair 5’dRP moiety as the cells lack the 5’dRP lysase function causing the cells to become hypersensitive to alkylating agents. Therefore 88 TAg Stable Y-LIZ DNA Polymerase-β Knockout Mouse Embryonic Fibroblast Cell Line is a valuable tool to evaluate the effects of the polymerase-β enzyme.

Propagation Requirements
Use of PriCoatTMT25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells inECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.
The base medium for this cell line is Prigrow III medium available at abm, Cat. No. TM003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999) to a final concentration of 10% and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.
