DescriptionTuberculosis (TB) remains a serious global health challenge despite numerous control programs enacted worldwide. The World Health Organization estimates that over 2 billion people are infected with TB and approximately 10 million new active cases are reported each year. Mycobacterium tuberculosis complex (MTbC) are a genetically related group of Mycobacterium species that causes TB in humans and includes M. tuberculosis, M. africanum, M. bovis, M. microti, M. canettii, M. caprae, M. pinnipedii, and M. mungi. Tuberculosis can have serious or fatal sequelae if left untreated and is readily transmissible, thus detection of active infection is important both for patient health management and for infection control measures. Molecular test approaches offer speed, sensitivity, and specificity advantages over classical test methods including Mantoux (skin) test, microscopic examination of sputum for acid-fast bacilli, or culture (6 to 8 weeks).
abm’s Tuberculosis (MTbC) Detection (qPCR) Kit offers a simple, sensitive molecular test system for the detection of MTbC DNA in sample material, utilizing a real-time PCR assay format designed to help minimize laboratory contamination. The kit contains a ready-to-use mix of customized qPCR reagents in paired (two-tube) strips.

Protocol OverviewIsolate DNA from sample(s) following decontamination and liquefaction steps as appropriate by sample type. abm recommends mechanical disruption methods with zirconia or glass beads, followed by silica based magnetic bead or spin-column purification.

Following DNA extraction, 8 µl aliquots of nucleic acid extract are added to each of the paired Control and Test reaction tubes and thermocycled in a qPCR machine. The Control tube serves as a reaction control by amplifying a human gene to ensure that sample collection, preparation, and qPCR reaction were performed properly. The Test tube performs a sensitive test for a genetic marker specific to MTbC as the tuberculosis detection assay.
