Supplementation may be required, most commonly with 5-10% fetal bovine serum (FBS) for cell culture use, and requires a 5-10% CO2 environment to maintain physiological pH, or be used as a base for development of a serum free medium.

EMEM is modified as follows:

With
• L-Glutamine
• Earle's Salts
• Non-essential Amino Acids
• 2.2 g/L Sodium Bicarbonate
