Storage of Adenovirus
At -80°C in 10 mM Tris (pH 8.0) plus 10% glycerol, the recombinant adenoviruses could be stable for several years. Alternatively, viral stock could be stored in 10mM Tris (pH 8.0) plus 4% sucrose for in vivo injection as it is more difficult to do in vivo injection with glycerol formulation. At 4°C, it can be stored for up to 2 weeks without significantly losing its titer.
Large Scale Production
For large-scale production of recombinant adenoviruses, it is recommended to infect 293 cells at 10 MOI, starting from a T25 flask and scale up to 50X T150 flasks. Total cell lysates containing recombinant adenovirus can be purified using either CsCl or filtration protocol. For more information, contact us at technical@abmGood.com
