Application

PBMC Isolation Protocol
- Prepare the Separation Medium
Bring the separation medium to room temperature and keep it protected from light.
- Add Separation Medium
Pipette 15 mL of the separation medium through the central hole of the built-in separation bracket. Ensure the medium level is above the bracket.
- Add Blood Sample
Gently add the anticoagulated whole blood (diluted if necessary) down the side of the tube, avoiding direct entry through the central hole to prevent mixing with the separation medium.
- Centrifugation
Centrifuge at 1200 ×g for 10 minutes at room temperature with the brake on. After centrifugation, the layers from top to bottom will be: plasma, PBMCs, separation medium, separation bracket, and erythrocytes/granulocytes.
- Collect PBMC Layer
Carefully pour off the top layer containing PBMCs and plasma into a new clean centrifuge tube or pipette out the PBMC layer.
- Wash PBMCs
Add phosphate-buffered saline (PBS) to the collected PBMCs and centrifuge at 250 ×g for 10 minutes at room temperature. Repeat this wash step once more.
- Resuspend Cells
After the final wash, resuspend the PBMCs in PBS or an appropriate culture medium for downstream applications.
