Lentiviral vector expressing the CRISPR adenine base editor (ABE) with GFP reporter. The construct combines nCas9 with a mutant version of TadA which enables precise A•T to G•C base pair conversions in DNA without introducing double-stranded breaks.

The key advantage of the CRISPR Adenine Base Editor (ABE) over regular Cas9 is that it enables precise A→G (T→C) base conversions without creating double-strand DNA breaks. This avoids the risks of random insertions/deletions from error-prone repair pathways, reduces off-target mutations, and allows efficient correction of point mutations that can cause many genetic diseases. In contrast, regular Cas9 relies on cleaving both strands of DNA and cellular repair, which is less predictable and less precise.
