The Cas9 nickase introduces single-strand breaks instead of double-strand breaks, offering higher specificity and lower off-target effects, especially when used in paired nickase configurations. In contrast, regular Cas9 creates DSBs, which are efficient for gene knockout but more prone to off-target mutations.
