Carbamoyl phosphate synthetase I (CPS I) and ornithine transcarbamoylase (OTC), key enzymes in the urea cycle, were introduced to CHO cells by transfection with calcium phosphate to enable permanent CPS I and OTC gene expression. This cell line is suitable for use in the study of the urea cycle. Expression of these enzymes results in decreased ammonium ion production, making these cells ideal for research regarding the affect of ammonia levels on cell growth and productivity.

Propagation Requirements

Use of PriCoatTM T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.

The base medium for this cell line is Prigrow VIII medium available at abm, Cat. No. TM018. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 10%, Penicillin/Streptomycin Solution (G255) to a final concentration of 1%, HT Media Supplement (50×) Hybri-Max (Sigma) to a final concentration of 1X, HEPES (Sigma) to a final concentration of 25 mM, N-Carbamyl-L-glutamic acid (Sigma) to a final concentration of 1 mM, and L-ornithine (Sigma) to a final concentration of 5 mM.
Change media every 2-3 days.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.
* Do not use heat-inactivated FBS for cell culture unless specified otherwise.

Preservation Protocol1. Freeze Medium: Prigrow VIII with 40% FBS and 10% DMSO.
