Luciferase is commonly used as a reporter to assess the transcriptional activity of a genetic construct with luciferase gene under the promoter of interest. Luciferase stable was generated using abm's pLenti-III-UbC-Luciferase lentivirus on U87MG/ΔEGFR Cells. The expression of luciferase has been confirmed with luciferase assay. All clones are puromycin (0.7ug/ml)-resistant. Luciferase stable U87MG/ΔEGFR cells can be a very useful cell line for non-invasive visualization in in vitro experiments.

Propagation Requirements

The base medium for this cell line is Prigrow III medium available at abm, Cat. No. TM003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999) to a final concentration of 10% and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%. Carbon dioxide (CO2): 5%, Temperature: 37.0°C.

Should I culture this cell line in the presence (0.7 ug/ml) or absence of puromycin in the cell culture media ?
This cell line should be kept under a selection pressure to ensure that the cells do not lose the stable luciferase expression. It is recommended to use 0.2-0.5ug/ml puromycin for daily culture.
Can you describe the U87MG/deltaEGFR cell line?
U87MG/deltaEGFR cell line expresses a truncated form of epidermal growth factor receptor (EGFR) - Δ(2-7) EGFR (in-frame deletion of 801bp in exons 2-7, also known as type III mutation). U87MG/deltaEGFR expresses EGFRvIII (mutant epidermal growth factor receptor). The method used to generate this cell line is via Retroviral vector. U87MG/deltaEGFR also confers a high malignancy to tumour cells.
Is the GFP or RFP signal localized within the cell?

What type of GFP or RFP do these cell lines express, and from which promoter?
The GFP is derived from copGFP (from Pontellina plumata) and the RFP is derived from mKate2 (from Entacmaea quadricolor). They are expressed from a proximal SV40 promoter, and a more distal CMV promoter may also drive expression.
