Probasco Cells (Luciferase Transduced)

Probasco cells were derived from a mixed breed dog that had spontaneous prostate cancer. Probasco cells were shown to metastasize to the appendicular and axial skeleton after injection into the left cardiac ventricle in mice and produce osteoblastic bone metastases. T8277 cells have been transduced with a luciferase reporter which allows for monitoring of tumor growth using bioluminescent imaging. The Probasco cell line is a valuable model to investigate the mechanisms of prostate cancer pathogenesis and osteoblastic bone metastases.

Propagation Requirements

Use of PriCoatTM T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.

The base medium for this cell line is Prigrow IV medium available at abm, Cat. No. TM004. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 10% and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.
* abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.

Preservation Protocol1. Freeze Medium: Complete growth medium with 20% FBS and 10% DMSO.
2. Storage Temperature: Liquid nitrogen vapour phase.

If frozen cells were received and not recovered in your lab under the exact, specified conditions (using recommended culture vessel, media, additional supplements, and atmospheric conditions), a live cell replacement is possible at a cost (plus shipping).
