Mannan-binding lectin-associated serine protease-1 (MASP-1) is a protein of the lectin pathway. Its similar in terms of structural features and substrate specificity to thrombin. Via its ability to induce fibrin clot formation along with interplay with several coagulation factors, MASP-1 has been shown to activate prothrombin. With the Prothrombin Mutant (R393Q) Stable Expressing HEK293 Cell Line, you can investigate in detail the mechanism of MASP-1 in mediating prothrombin activation.

Propagation Requirements

Use of PriCoatTM T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.

The base medium for this cell line is Prigrow III medium available at abm, Cat. No. TM003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 5%, sodium pyruvate (TM057) to a final concentration of 1 mM, non-essential amino acids to a final concentration of 1X, HEPES to a final concentration of 10 mM.
Change media every 2-3 days.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.
* Do not use heat-inactivated FBS for cell culture unless specified otherwise.

Preservation Protocol1. Freeze Medium: Complete growth medium with 10% FBS and 5% DMSO.
2. Storage Temperature: Liquid nitrogen vapour phase.
