| What is the difference between saCas9 and spCas9? | |
|
spCas9 is larger and recognizes the shorter PAM NGG, making it the most widely used and versatile Cas9. saCas9 is smaller, making it ideal for usage in AAV vectors, and recognizes a longer PAM NNGRRT, which provides higher specificity but fewer targetable sites. In practice, spCas9 is the standard for in vitro work, while saCas9 is often preferred for in vivo therapeutic delivery.

The main advantage of saCas9 over spCas9 is its smaller size, which allows it to be packaged together with its sgRNA into a single AAV vector for efficient in vivo delivery. This makes saCas9 especially valuable for therapeutic applications where delivery capacity is limited.

| Do you offer this vector/virus with a fluorescent marker? | |
|
No. AAV has a limited cargo size of ~4.4kb thus the addition of a promoter, reporter and polyA signal would exceed 4.4kb and the construct would not be able to be viral packaged.

| Do I need to provide my own sgRNA? | |
|
Yes, this product only expresses the Cas9 or Cas-related protein. We offer genome-wide sgRNA only expressing vectors for CRISPR gene knockout (spCas9 and saCas9) and CRISPR gene activation and repression which are available through the Customize Vector option.
