DescriptionThe Stable PML Knockout Hep-2 Cell Line was generated from HEp-2 cells by transfection of CRISPR/Cas9 targeting exon 1 of promyelocytic leukemia protein (PML). PML is a component of nuclear domain 10 (ND10) and an antiviral effector of IFN- β. In the absence of PML, the antiviral effects of IFN- β are significantly reduced. This makes the Stable PML Knockout Hep-2 Cell Line a useful model in antiviral treatments and experiments. The PML gene is also associated with a wide range of important cellular processes including tumor suppression, transcriptional regulation, apoptosis, senescence, and DNA damage response which can all be further studied using this cell line.

Propagation Requirements
Use of PriCoatTMT25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells inECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.
The base medium for this cell line is Prigrow III medium available at abm, Cat. No. TM003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999) to a final concentration of 10% and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.
