Descriptionc-Kit or stem cell factor (SCF) receptor is a type III tyrosine kinase receptor that has a major role in the hematopoietic system. Gain-of-function mutations of the receptor have been observed in malignancies such as myeloid leukemia, gastrointestinal stromal tumors, and testicular carcinoma. While loss-of-function mutations were found in defects relating to hematopoiesis, melanogenesis, and gametogenesis. The c-Kit/V560D-expressing Ba/F3 cell line (with GNNK- isoform) has a substitution of the valine residue at position 560 to aspartic acid. The cells are resistant to Imatinib (Gleevec), an inhibitor of the c-Kit juxtamembrane mutations. Together with the wild-type c-Kit stable Ba/F3 cells (Cat. No. T3041 and T3042), these cell lines are suitable for studies in drug discovery for mutations in the c-Kit receptor which are associated to oncogenic and other diseases.

Propagation RequirementsThe base medium for this cell line is Prigrow II medium available at abm, Cat. No. TM002. To make the completed growth medium, add the following components to the base medium: 10 ng/ml recombinant murine IL-3, fetal bovine serum (TM999) to a final concentration of 10%, Penicillin/Streptomycin Solution (G255) to a final concentration of 1%, and 1.2 μg/ml puromycin.

      What is the biosafety level for these cells?
      In accordance to the U.S. Public Health Service Guidelines, the designation of Ba/F3, a murine Pro B cell line, falls under laboratory biosafety level 1 (BSL-1). Biosafety Level 1 is suitable for work involving well-characterized agents not known to cause disease in humans and present minimal potential hazard to the laboratory personnel and the environment. It is the responsibility of the end user to ensure that their facility comply with biosafety regulations for their own country. The regulation guidelines include, but not limited to: Enforcement of standard laboratory practices:
      • No eating, drinking and smoking permitted in laboratory areas
      • No mouth pipetting
      • Safe handling of sharps, such as needles, pipettes, broken glassware
