Description

Fusing a kinase domain to a homodimerization domain such as the PNT domain of ETV6 can result in activated kinase. The FLT3 Stable Ba/F3 Cell Line expresses activated FLT3 tyrosine kinase that is linked to a GFP reporter (GFP-ETV6-FLT3 in MSCV-puro vector). Ba/F3 cells expressing activated tyrosine kinase become completely dependent on the kinase activity for survival and as a result, the cells can be propagated even in the absence of IL-3.

FLT3 belongs to the platelet-derived growth factor (PDGF) family of receptor tyrosine kinases consisting of proteins which bind to platelet-derived growth factors. These proteins are heavily involved in embryonic development, acting as early as gastrulation; they assist in proper organ formation as well as hematopoiesis. Upon binding of the fms-related tyrosine kinase 3 ligand, the activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow. Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia.

Propagation Requirements

The base medium for this cell line is Prigrow II medium available at abm, Cat. No. TM002. To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999) to a final concentration of 10%, Penicillin/Streptomycin Solution (G255) to a final concentration of 1%, and Recombinant Mouse IL3 (Z200145) to a final concentration of 5 ng/mL. May be grown in the presence of 1?μg/ml puromycin, but it is not required for optimal cell growth.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.

After thawing them, is FLT3 expression still high at the cell surface?
The FLT3 kinase domain is expressed in the cells which is determined by Western blot. Therefore, the cells express the kinase domain but this does not specify the expression to be at the cell surface.
As FLT3 tyrosine kinase is linked to a GFP reporter, does the binding of human FLT3-Ligand induce GFP expression or is it stably expressed as a tag or IRES/2A co-expression system?
The FLT3 kinase is fused with the PNT domain of the ETV6 to result in an activated kinase, and GFP reporter is linked to the kinase. This GFP is a tag for FACS and does not reflect expression level of the protein.
What is the biosafety level for these cells?
In accordance to the U.S. Public Health Service Guidelines, the designation of Ba/F3, a murine Pro B cell line, falls under laboratory biosafety level 1 (BSL-1). Biosafety Level 1 is suitable for work involving well-characterized agents not known to cause disease in humans and present minimal potential hazard to the laboratory personnel and the environment. It is the responsibility of the end user to ensure that their facility comply with biosafety regulations for their own country. The regulation guidelines include, but not limited to: Enforcement of standard laboratory practices:
  • No eating, drinking and smoking permitted in laboratory areas
