Growth Conditions

PriCoat™ T25 Flasks (G299) are recommended for optimal cell culture. The optimal media formulations and growth conditions are detailed below.

Thawing Medium: PrimGrow T Xpan™ Basal Medium (TM182-B) + 10% FBS + 10 µl ROCK Inhibitor Y-27632 (TM131+ 1% Penicillin/Streptomycin Solution (G255).

Activation Medium: PrimGrow T Xpan™ Basal Medium (TM182-B) with T Xpan™ Cell Activator (CD3/CD28/CD2) (TM139) at 2 µl per 1 ml of basal medium.

Maintenance/Expansion Medium: PrimGrow T Xpan™ Medium Kit  (TM182) + 1% Penicillin/Streptomycin Solution (G255).  

Incubate cells at 37.0°C, 5% CO.  

Note: Cells must be activated immediately after thawing before use in downstream applications.

Unpacking and Storage Instructions

1. Visually examine the packaging containers for signs of leakage or breakage.

2. Immediately transfer frozen cells from dry ice packaging to a temperature below -130°C, preferably in liquid nitrogen vapor phase storage, until ready for use.

To ensure the highest level of viability, thaw the vial and initiate culture as soon as possible upon receipt. If continued storage is desired, the vial should only be stored below -130°C or in liquid nitrogen vapor phase. Do not store at -70°C, as it will result in loss of viability.

Thawing Protocol

1. Thaw cells quickly in a 37°C water bath while agitating gently (maximum 2 minutes). The vial cap should be kept above the water level to minimize the risk of contamination.

2. Decontaminate the vial by spraying and wiping the exterior of the vial with 70% ethanol. From this point onwards, all operations should be strictly carried out inside a biological safety cabinet using aseptic conditions.

3. Transfer the cell suspension into a 15ml sterile conical tube containing 10ml of pre-warmed Thawing Medium. Centrifuge cells at 125 xg for 5-7 minutes.

4. Carefully aspirate the supernatant, leaving a thin layer of medium above the pellet. Resuspend the cell pellet in Activation Medium and transfer the entire volume into a PriCoat™ T25 flask (G299). Example: 5 ml basal media + 10 µl T Xpan™ Cell Activator.

5. Incubate the cells for 24 hours.

6. Check for cluster formation, typically visible between 24–48 hours. Once clusters are present, activated cells are suitable for downstream applications.

7. After confirming activation, replace the activation medium with Maintenance/Expansion Medium. Do not allow cell density to exceed 3 × 10⁶ cells/ml. Subculture accordingly.

8. For ongoing expansion, re-stimulate cultures every 7–14 days using 2 µl of T Xpan™ Cell Activator (TM139) per 1 ml of cell suspension.


Subculture Protocol

Do not allow cell density to exceed exceeds 3 × 10⁶ cells/ml, subculture cells as per the instructions below. Confluent cultures may be transferred to T25 or T75 culture flasks which can be split to additional flasks. 


1. Simply add fresh complete media directly to the culture flask. 


or,


2. Remove half of the cell suspension and transfer to a new culture vessel; add additional fresh complete media to both culture vessels.

3. Incubate the cells at the recommended conditions.

4. Continue to monitor cell growth (cell clumps should continue to grow and become visible in the culture flask). Add fresh complete growth media every 3 days.


5. Rapid proliferation typically continues for up to 14 days, during which cells should be passaged every 1–2 days. Afterward, they return to a more stable growth phase. 

6. If additional expansion is needed after day 14, a second activation (repeat the 24-well setup) may be performed only when cells are healthy. However, avoid repeated stimulations — more than 3–5 rounds can induce T cell exhaustion.


Note: Do not centrifuge cells during subculture procedures.

We recommend using serum-free CryoGuard™  Freezing Media (TM078).

Immortalization Method

Serial passaging and retroviral transduction with retroviruses carrying the SV40 gene.
