DescriptionU251MG cells were stably transfected with the rtTA expression vector to generate the U251MG Stably Expressing Tet-on Cell Line. It can be used in vitro to stably transfect gene of interest cloned in tetracycline-responsive expression vector (i.e. pTRE2 with Hygromycin selection) and pick clones. With the addition of 1 µg/ml doxycycline, end users can test individually for induced gene expression under the tet-regulated promoter.

Propagation Requirements
Use of PriCoatTMT25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells inECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.
The base medium for this cell line is Prigrow IV medium available at abm, Cat. No. TM004. To make the complete growth medium, add the following components to the base medium: tet-tested fetal bovine serum to a final concentration of 10%, 1% L-glutamine (G275), 200 ug/ml G418 (G271), and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%.
Carbon dioxide (CO2): 5%, Temperature: 37.0°C.

After successful transfection of gene of interest in tetracycline-responsive vector and picking colonies, 1 μg/ml doxycycline may be used for induction of expression.
